Tissue Micro Arrays. TMA are constructed by using a needle to biopsy a standard histologic sections and placing the core in an array on a recipient paraffin block. Hundreds or thousands of tissue cores are arranged on a single slide and then analyzed by a single immuno-staining experiment or in situ hybridization reaction. TMAs built are highly representative of donor tissues with confirmed pathological diagnosis.
Salient Features of TMAs over single sections:

TMA are constructed by using a biopsy needle from the donor blocks then  placing the core in an array on a recipient paraffin block.

TMAs conserve tissue resources and experimental reagents
Improve internal experimental controls
Increase sample numbers per experiment
Can be used for large-scale, massively parallel in situ analysis.
Triesta offers custom made TMAs with various tissue types and variable replicates depending on the requirement.

Biomarker Analysis.
Triesta's biomarker research is focused on the identification, measurement and clinical validation of novel DNA, RNA and protein-based targets and biomarkers at the level of gene expression and epigenetic changes.

Triesta offers its capabilities to partners and customers as a service or through research collaboration with shared risks and rewards:

1 Biomarker discovery
2 Clinical validation / replication studies
3 Assay development and standardization
Extraction of DNA and RNA. Triesta offers variety of methods to extract top quality nucleic acids from various types of biological material not limited to FFPE blocks, fresh frozen tissue, blood, yeast, bacteria and cell culture.
Cell-culture: Triesta’s well established cell culture facilities are well equipped to maintain cell lines, primary cultures and transfection assays.
Mutational Analysis. We offer standardized methods of mutation detection either by direct DNA sequencing or ARMS PCR. Mutation of EGFR, K-ras and p53, to mention a few, are offered for diagnostic and research work.
FISH. EGFR and Her-2/neu gene amplification are being studied by Fluorescent in situ hybridization (FISH).
Cytogenetics. Conventional karyotyping, cell cultures, FISH on metaphase, solid tumor cytogenetics for diagnosis as well as for research.
Flow cytometry based assays. Triesta has the expertise for various flow cytometer based assays, not limited to immunophenotyping, DNA ploidy and drug resistance
LCM: The MMI SmartCut® system at Triesta is a tool to isolate clean and pure population of cells. This is achieved by dissecting the areas of interest with a UV-laser while maintaining morphology, ensuring quality of the source material and contamination free collections for subsequent downstream analysis.

Applications:
Differential analysis of genomic and proteomic expression of any tissue type.
Single cell isolation: Single cells can be easily identified and isolated from tissue sections, fine needle aspiration smears, blood and cell cultures
Live cells can be captured from a suspension and can be re-cultured without any adverse cellular effects.
